Methodology/PrincipalFindings:Inthisstudy,weinvestigatedeffectsofAngIIandAng-(1-7)onvascularsmoothmuscle cell(SMC)proliferationandmigration,whicharecriticalintheformationofatheroscleroticlesions.TreatmentwithAngII resultedinanincreaseofSMCproliferation,whereasAng-(1-7)alonehadnoeffects.However,preincubationwithAng-(1-7) inhibitedAngII-inducedSMCproliferation.AngIIpromotedSMCmigration,andthiseffectwasabolishedbypretreatment withAng-(1-7).ThestimulatoryeffectsofAngIIonSMCproliferationandmigrationwereblockedbytheAngIIreceptor antagonistlorsartan,whiletheinhibitoryeffectsofAng-(1-7)wereabolishedbytheAng-(1-7)receptorantagonistA-799. Ang II treatment caused activation of ERK1/2 mediated signaling, and this was inhibited by preincubation of SMCs with Ang-(1-7). Conclusion:TheseresultssuggestthatAng-(1-7)inhibitsAngII-inducedSMCproliferationandmigration,atleastinpart, throughnegativemodulationofAngIIinducedERK1/2activity.
