The aim of this study was to evaluate multiplexed bead-based immunoassay of multiple ovarian cancer-associated biomarkers such as transthyretin and apolipoproteinA1,togetherwithCA125,toimprovetheidentificationandevaluationofprognosisofovariancancer.We measured the serum levels of CA125, transthyretin, and apolipoprotein A1 from the serum of 61 healthy individuals, 84 patientswithbenignovariandisease,and118patientswithovariancancerusingamultiplexliquidassaysystem,Luminex 100.Theresultswerethenanalyzedaccordingtohealthyand/orbenignversusovariancancersubjects.WhenCA125was combinedwiththeotherbiomarkers,theoverallsensitivityandspecificityweresignificantlyimprovedintheROCcurve, whichshowed95%and97%sensitivityandspecificity,respectively.At95%specificityforallstagesthesensitivityincreased to95.5%comparedto67%forCA125alone.ForstageI+II,thesensitivityincreasedfrom30%forCA125aloneto93.9%.For stageIII+IV,thecorrespondingvalueswere96.5%and91.6%,respectively. Also,thethreebiomarkersweresufficientfor maximumseparationbetweennoncancer(healthyplusbenigngroup)andstageI+IIorallstages(I2IV)ofdisease.Thenew combinationoftransthyretin,andapolipoproteinA1withCA125improvedboththesensitivityandthespecificityofovarian cancerdiagnosiscomparedwiththoseofindividualbiomarkers.Thesefindingssuggestthebenefitofthecombinationof thesemarkersforthediagnosisofovariancancer.
