Abstract
Push–pullnetworks,inwhichtwoantagonisticenzymescontroltheactivityofamessengerprotein,areubiquitousinsignal
transductionpathways.AclassicalexampleisthechemotaxissystemofthebacteriumEscherichiacoli,inwhichthekinaseCheA
andthephosphataseCheZregulatethephosphorylationlevelofthemessengerproteinCheY.Recentexperimentssuggestthat
boththekinaseandthephosphatasearelocalizedatthereceptorcluster,andVakninandBergrecentlydemonstratedthatthe
spatialdistributionofthephosphatasecanmarkedlyaffectthedose–responsecurves.Weargue,usingmathematicalmodeling,
thatthecanonicalmodelofthechemotaxisnetworkcannotexplaintheexperimentalobservationsofVakninandBerg.We
presentanewmodel,inwhichasmallfractionofthephosphataseislocalizedatthereceptorcluster,whiletheremainderfreely
diffusesinthecytoplasm;moreover,thephosphataseattheclusterhasahigherbindingaffinityforthemessengerproteinand
ahighercatalyticactivitythanthephosphataseinthecytoplasm.Thismodelisconsistentwithalargebodyofexperimental
dataandcanexplainmanyoftheexperimentalobservationsofVakninandBerg.Moregenerally,thecombinationofdifferential
affinity and catalytic activity provides a generic mechanism for amplifying signals that could be exploited in other two-
componentsignalingsystems.Ifthismodeliscorrect,thenanumberofrecentmodelingstudies,whichaimtoexplainthe
chemotacticgainintermsoftheactivityofthereceptorcluster,shouldbereconsidered.
