Abstract
Zebrafish express in their retina a higher number of guanylate cyclase-activating proteins (zGCAPs) than mammalians
pointingtomorecomplexguanylatecyclasesignalingsystems.AllsixzGCAPisoformsshowdistinctandpartialoverlapping
expression profiles in rods and cones. We determined critical Ca -dependent parameters of their functional properties

usingpurifiedzGCAPsafterheterologousexpressioninE.coli.Isoforms1–4werestrong,5and7wereweakactivatorsof
membrane bound guanylate cyclase. They further displayed different Ca -sensitivities of guanylate cyclase activation,

whichishalfmaximaleitheratafreeCa around30nM(zGCAP1,2and3)oraround400nM(zGCAP4,5and7).Zebrafish
GCAPisoformsshowedalsodifferencesintheirCa /Mg -dependentconformationalchangesandintheCa -dependent
monomer-dimer equilibrium. Direct Ca -binding revealed that allzGCAPs bound at least three Ca . Thecorresponding

apparentaffinityconstantsreflectbindingofCa2+withhigh(#100nM),medium(0.1–5mM)and/orlow($5mM)affinity,
butwereuniqueforeachzGCAPisoform.OurdataindicateaCa -sensorsysteminzebrafishrodandconecellssupporting
aCa -relaymodelofdifferentialzGCAPoperationinthesecells.
