In the present study, we comprehensively compared the biological featuresandgeneexpressionprofileofBM-MSCsbetweenAApatientsandhealthyvolunteers.Incomparisonwithhealthy controls,BM-MSCsfromAApatientsshowedaberrantmorphology,decreasedproliferationandclonogenicpotentialand increasedapoptosis.BM-MSCsfromAApatientsweresusceptibletobeinducedtodifferentiateintoadipocytesbutmore difficulttodifferentiateintoosteoblasts.Consistentwithabnormalbiologicalfeatures,alargenumberofgenesimplicated incellcycle,celldivision,proliferation,chemotaxisandhematopoieticcelllineageshowedmarkedlydecreasedexpression inBM-MSCsfromAApatients.Conversely,morerelatedgeneswithapoptosis,adipogenesisandimmuneresponseshowed increased expression in BM-MSCs from AA patients. The gene expression profile of BM-MSCs further confirmed the abnormalbiologicalpropertiesandprovidedsignificantevidenceforthepossiblemechanismofthedestructionofthebone marrowmicroenvironmentinAA.
