ThehomologousgeneinHHV-6Bisdr6.Sincep53isderegulatedinbothHHV-6Aand-6B,wecharacterizedtheexpression ofdr6mRNAandthelocalizationofthetranslatedproteinduringHHV-6BinfectionofHCT116cells.ExpressionofmRNA fromdr6wasinhibitedbycycloheximideandpartlybyphosphonoaceticacid,aknowncharacteristicofherpesvirusearly/ lategenes.DR6couldbedetectedasanuclearproteinat24hpiandaccumulatedtohighlevelsat48and72hpi.DR6 locatedindotsresemblingviralreplicationcompartments.Furthermore,anovelinteractionbetweenDR6andtheviralDNA processivity factor, p41, could be detected by confocal microscopy and by co-immunoprecipitation analysis. In contrast, DR6andp53werefoundatdistinctsubcellularlocations.Together,ourdataimplyanovelfunctionofDR6duringHHV-6B replication.
