DistinctPatternsofHIV-1EvolutionwithinMetastatic TissuesinPatientswithNon-HodgkinsLymphoma MarcoSalemi1*,SusannaL.Lamers2,LeanneC.Huysentruyt3,DerekGalligan3,RebeccaR.Gray1 ,Alanna Morris3,MichaelS.McGrath3* 1Department of Pathology Immunology and Laboratory Medicine, University of Florida, Gainesville, Florida, United States of America, 2BioInfoExperts, Thibodaux, Louisiana,UnitedStatesofAmerica,3DepartmentofMedicine,HematologyandOncology,UniversityofCaliforniaSanFrancisco,SanFrancisco,California,UnitedStates ofAmerica Abstract Despitehighlyactiveantiretroviraltherapy(HAART),AIDSrelatedlymphoma(ARL)occursatasignificantlyhigherratein patientsinfectedwiththeHumanImmunodeficiencyVirus(HIV)thaninthegeneralpopulation.HIV-infectedmacrophages areaknownviral reservoir and havebeenshown tohavelymphomagenic potential in SCID mice; therefore, there is an interestindeterminingifaviralcomponenttolymphomagenesisalsoexists.WesequencedHIV-1envelopegp120clones obtained post mortem from several tumor and non-tumor tissues of two patients who died with AIDS-related Non- Hodgkin’slymphoma(ARL-NH).Similarresultswerefoundinbothpatients:1)high-resolutionphylogeneticanalysisshowed asignificantdegreeofcompartmentalizationbetweenlymphomaandnon-lymphomaviralsub-populationswhileviralsub- populationsfromlymphnodesappearedtobeintermixedwithinsequencesfromtumorandnon-tumortissues,2)a100- foldincreaseintheeffectiveHIVpopulationsizeintumorversusnon-tumortissueswasassociatedwiththeemergenceof lymphadenopathyandaggressivemetastaticARL,and3)HIVgeneflowamonglymphnodes,normalandmetastatictissues wasnon-random.Thedifferentpopulationdynamicsbetweenthevirusesfoundintumorsversusthenon-tumorassociated virusessuggestthatthereisasignificantrelationshipbetweenHIVevolutionandlymphomapathogenesis.Moreover,the studyindicatesthatHIVcouldbeusedasaneffectivemarkertostudytheoriginanddisseminationoflymphomasinvivo.
