MutationsoftheH3K27methyltransferasegenes,ARABIDOPSISTRITHORAX-RELATEDPROTEIN5(ATXR5)andATXR6,resultin re-replication(repeatedoriginfiringwithinthesamecellcycle).HereweshowthatmutationsthatreduceDNAmethylation acttosuppressthere-replicationphenotypeofatxr5atxr6mutants.ThissuggeststhatDNAmethylation,amarkenrichedat thesameheterochromaticregionsthatre-replicateinatxr5/6mutants,isrequiredforaberrantre-replication.Incontrast, RNAsequencinganalysessuggestthatATXR5/6andDNAmethylationcooperativelytranscriptionallysilencetransposable elements(TEs).HenceourresultssuggestacomplexrelationshipbetweenATXR5/6andDNAmethylationintheregulation ofDNAreplicationandtranscriptionofTEs.
