DownregulationofRPL6bysiRNAInhibitsProliferation andCellCycleProgressionofHumanGastricCancerCell Lines QiongWu1.,YawenGou1,2.,QiaominWang2,HaifengJin1,LinaCui1,YongguoZhang1,LijieHe1 ,Jingbo Wang1,YongzhanNie1,YongquanShi1*,DaimingFan1* 1StateKeyLaboratoryofCancerBiology,XijingHospitalofDigestiveDiseases,theFourthMilitaryMedicalUniversity,Xi’an,Shaanxi,China,2DepartmentofDigestive Diseases,TheAffiliatedProvincialHospitalofAnhuiMedicalUniversity,Hefei,AnhuiProvince,China Abstract OurpreviousstudyrevealedthathumanribosomalproteinL6(RPL6)wasup-regulatedinmultidrug-resistantgastriccancer cellsandover-expressionofRPL6couldprotectgastriccancerfromdrug-inducedapoptosis.Itwasfurtherdemonstrated that up-regulation of RPL6 accelerated growth and enhanced in vitro colony forming ability of GES cells while down- regulationofRPL6exhibitedtheoppositeresults.ThepresentstudywasdesignedtoinvestigatethepotentialroleofRPL6 in therapy of gastric cancer for clinic. The expression of RPL6 and cyclin E in gastric cancer tissues and normal gastric mucosawasevaluatedbyimmunohistochemisty.ItwasfoundthatRPL6andcyclinEwereexpressedatahigherlevelin gastriccancertissuesthanthatinnormalgastricmucosaandthetwowerecorrelativeingastriccancer.Survivaltimeof postoperativepatientswasanalyzedbyKaplan-MeieranalysisanditwasfoundthatpatientswithRPL6positiveexpression showedshortersurvivaltimethanpatientsthatwithRPL6negativeexpression.RPL6wasthengeneticallydown-regulated ingastriccancerSGC7901andAGScelllinesbysiRNA.Itwasdemonstratedthatdown-regulationofRPL6reducedcolony formingabilityofgastriccancercellsinvitroandreducedcellgrowthinvivo.Moreover,down-regulationofRPL6could suppressG1toSphasetransitioninthesecells.Further,weevidencedthatRPL6siRNAdown-regulatedcyclinEexpression inSGC7901andAGScells.Takentogether,thesedatasuggestedthatRPL6wasover-expressedinhumangastrictissuesand caused poorprognosis.
