The frequency of the amino acid substitution D222G in the hemagglutinin (HA) of 2009 H1N1 viruses isolated from severe but not mild human cases represents the first molecular marker associated with enhanced disease. To assess the relative contribution of this substitutioninviruspathogenesis,transmission,andtropism,weintroducedD222Gbyreversegeneticsinthewild-typeHA ofthe2009H1N1virus,A/California/04/09(CA/04).Adose-dependentglycanarrayanalysiswiththeD222Gvirusshoweda modestreductioninthebindingaviditytohuman-like(a2-6sialylatedglycan)receptorsandanincreaseinthebindingto avian-like(a2-3sialylatedglycan)receptorsincomparisonwithwild-typevirus.Intheferretpathogenesismodel,theD222G mutantviruswasfoundtobesimilartowild-typeCA/04viruswithrespecttolethargy,weightlossandreplicationefficiency intheupperandlowerrespiratorytract.Moreover,basedonviraldetection,therespiratorydroplettransmissionproperties ofthesetwoviruseswerefoundtobesimilar.TheD222Gvirusfailedtoproductivelyinfectmiceinoculatedbytheocular route,butexhibitedgreaterviralreplicationandweightlossthanwild-typeCA/04virusinmiceinoculatedbytheintranasal route.I
