In general, studies of codonusagebias(CUB)assumethatcodonswithhighertRNAabundancehavelowermissenseerrorrates.Usingamodelof proteintranslationbasedontRNAcompetitionandintra-ribosomalkinetics,weshowthatthisassumptioncanbeviolated whentRNAabundancesarepositivelycorrelatedacrossthegeneticcode.ExaminingthedistributionoftRNAabundances across73bacterialgenomesfrom20differentgenera,wefindaconsistentpositivecorrelationbetweentRNAabundances acrossthegeneticcode.Thisworkchallengesoneofthefundamentalassumptionsmadeinover30yearsofresearchon CUBthatcodonswithhighertRNAabundanceshavelowermissenseerrorratesandthatmissenseerrorsaretheprimary selectiveforceresponsibleforCUB.
