We identified andcharacterized the calcineurin genes PsCNA1andPsCNB1inPst.PhylogeneticanalysesindicatethatPsCNA1andPsCNB1formacalcium/calmodulinregulated protein phosphatase belonging to the calcineurin heterodimers composed of subunits A and B. Quantitative RT-PCR analysesrevealedthatbothPsCNA1andPsCNB1expressionreachedtheirmaximuminthestageofhaustoriumformation, whichisonedayafterinoculation.Usingbarelystripemosaicvirus(BSMV)asatransientexpressionvectorinwheat,the expression of PsCNA1 and PsCNB1 in Pst was suppressed, leading to slower extension of fungal hyphae and reduced productionofurediospores.Theimmune-suppressivedrugscyclosporinAandFK506markedlyreducedthegermination ratesofurediospores,andwhengerminationdidoccur,morethantwogermtubeswereproduced.Theseresultssuggest thatthecalcineurinsignalingpathwayparticipatesinstriperustmorphogeneticdifferentiation,especiallytheformationof haustoriaduringtheearlystageofinfectionandduringtheproductionofurediospores.ThereforePsCNA1andPsCNB1can beconsideredimportantpathogenicitygenesinvolvedinthewheat-Pstinteraction.
