FunctionalConservationoftheDrosophilagooseberry GeneandItsEvolutionaryAlleles WeiLiu1,3,LeiXue2,3 * 1College of Veterinary Medicine, Northwest Agriculture Forest University, Yangling, Shaanxi, China, 2School of Life Science and Technology, Tongji University, Shanghai,China,3InstituteforMolecularBiology,UniversityofZu¨rich,Zurich,Switzerland Abstract TheDrosophilaPaxgenegooseberry(gsb)isrequiredfordevelopmentofthelarvalcuticleandCNS,survivaltoadulthood, andmalefertility.Thesefunctionscanberescued in gsbmutantsbytwo gsb evolutionaryalleles, gsb-Prdandgsb-Pax3, which express the Drosophila Paired and mouse Pax3 proteins under the control of gooseberry cis-regulatory region. Therefore,bothPairedandPax3proteinshaveconservedalltheGsbfunctionsthatarerequiredforsurvivalofembryosto fertileadults,despitethedivergentprimarysequencesintheirC-terminalhalves.Asgsb-Prdandgsb-Pax3uncoveragsb functioninvolvedinmalefertility,constructionofevolutionaryallelesmayprovideapowerfulstrategytodissecthitherto unknown gene functions. Our results provide further evidence for the essential role of cis-regulatory regions in the functionaldiversificationofduplicatedgenesduringevolution.
