PhaeobactergallaeciensisReducesVibrioanguillarumin CulturesofMicroalgaeandRotifers,andPrevents VibriosisinCodLarvae PaulW.D’Alvise1,SirilLilleb?2,MariaJ.Prol-Garcia1,HeidrunI.Wergeland2,KristianF.Nielsen3, ?ivindBergh4,LoneGram1* 1NationalFoodInstitute,TechnicalUniversityofDenmark,Lyngby,Denmark,2DepartmentofBiology,UniversityofBergen,Bergen,Norway,3DepartmentofSystems Biology,CenterforMicrobiologicalBiotechnology,TechnicalUniversityofDenmark,Lyngby,Denmark,4InstituteofMarineResearch,Bergen,Norway Abstract Phaeobactergallaeciensiscanantagonizefish-pathogenicbacteriainvitro,andthepurposeofthisstudywastoevaluatethe organismasaprobiontformarinefishlarvaeandtheirfeedcultures.Aninvivomechanismofactionoftheantagonistic probioticbacteriumissuggestedusinganon-antagonisticmutant.P.gallaeciensiswasreadilyestablishedinaxeniccultures ofthetwomicroalgaeTetraselmissuecicaandNannochloropsisoculata,andoftherotiferBrachionusplicatilis.P.gallaeciensis reacheddensitiesof107cfu/mlanddidnotadverselyaffectgrowthofalgaeorrotifers.Vibrioanguillarumwassignificantly reducedbywild-typeP.gallaeciensis,whenintroducedintothesecultures.AP.gallaeciensismutantthatdidnotproduce theantibacterialcompoundtropodithieticacid(TDA)didnotreduceV.anguillarumnumbers,suggestingthatproductionof theantibacterialcompoundisimportantfortheantagonisticpropertiesofP.gallaeciensis.TheabilityofP.gallaeciensisto protectfishlarvaefromvibriosiswasdeterminedinabathchallengeexperimentusingamultidishsystemwith1larvaper well.Unchallengedlarvaereached40%accumulatedmortalitywhichincreasedto100%wheninfectedwithV.anguillarum. P.gallaeciensisreducedthemortalityofchallengedcodlarvae(Gadusmorhua)to10%,significantlybelowthelevelsofboth thechallengedandtheunchallengedlarvae.TheTDAmutantreducedmortalityofthecodlarvaeinsomeofthereplicates, althoughtoamuchlesserextentthanthewildtype.ItisconcludedthatP.gallaeciensisisapromisingprobiontinmarine larvicultureandthatTDAproductionlikelycontributestoitsprobioticeffect.
