Methods:Post-hocanalysisofa13-weekrandomized,double-blindplacebo-controlledstudyofOROSmethylphenidate (MPH)54and72mg/day.PrimaryefficacyvariablewastheConners’AdultADHDRatingScale–ScreeningVersion (CAARS:O-SV).Dailyadherencewascalculatedasaveragedailyadherence(100×capsulestaken/2),withoverall adherencecalculatedastheaveragedailyadherence.Predictorsofadherencewereassessedusingmixed-effectslogistic regression.DescriptivestatisticsweregeneratedforchangeinCAARS:O-SVscoreforadherent(95%adherence)and non-adherentsubjects.Predictorsofchangewereanalyzedusingamixedmodel. Results:SubjectswereallocatedtoOROSMPH(54mg,n=87;72mg,n=92)orplacebo(n=97).Meanadherence was92.6%and93.3%(OROSMPH54and72mg/day,respectively),versus97.5%(placebo).Adherencewashigherand lessvariableincompleters.Factorssignificantlyassociatedwithnon-adherenceincludedfemalesex,shortertimesince ADHDdiagnosis,highereducationlevel(completionofuniversity)andscoreontheDrugUseScreeningInventory psychiatricdisorderssubscale.ImprovementsfrombaselineinCAARS:O-SVscorewerenumericallygreaterinsubjects definedasadherentthaninthosewhowerenon-adherent.SignificantpredictorsofCAARS:O-SVchangeinpatients whocompletedthestudyincludedpercentageadherenceuptothepointofassessment(p0.0001),baselinescore (p0.0001)andfamilyhistoryofADHD(p=0.0003).
