PreliminaryInsightsintothePhylogeographyofSix AquaticHyphomyceteSpecies SofiaDuarte1*,SahadevanSeena1,FelixBa¨rlocher2,FernandaCa′ssio1,Cla′udiaPascoal1 1CentreofMolecularandEnvironmentalBiology (CBMA),DepartmentofBiology,UniversityofMinho, CampusdeGualtar, Braga,Portugal,2DepartmentofBiology, MountAllisonUniversity,Sackville,NewBrunswick,Canada Abstract Aquatichyphomycetesoccurworldwideonawiderangeofplantsubstratesdecomposinginfreshwaters,andareknownto playakeyroleinorganicmatterturnover.Thepresumedworldwidedistributionofmanyaquatichyphomycetespecieshas beenbasedonmorphology-basedtaxonomyandidentification,whichmayoverlookcrypticspecies,andmaskglobal-scale biogeographicalpatterns.ThismightbecircumventedbyusingDNAsequencedata.Theinternaltranscribedspacer(ITS) region from rDNA was recently designated as the most suitable barcode for fungal identification. In this study, we generatedITSbarcodesof130isolatesbelongingto6aquatichyphomycetespecies(Anguillosporafiliformis,Flagellospora penicillioides,Geniculosporagrandis,Lunulosporacurvula,TetrachaetumelegansandTricladiumchaetocladium),andcollected fromstreamsofSouthwestEurope(86isolates)andEastAustralia(44isolates).EuropeanandAustralianpopulationsof4 species(A.filiformis,F.penicillioides,G.grandisandT.elegans)groupedintodifferentclades,andmoleculardiversityindices supportedsignificantdifferentiation.Continentsdidnotsharehaplotypes,exceptforT.chaetocladium.Overallresultsshow substantialpopulationdiversityforalltestedspeciesandsuggeststhatthebiogeographyofaquatichyphomycetesmaybe species-specific.
