Methods:PrevalenceofC.trachomatisinfectionamongwomenvisitinggynaecologydepartmentofHinduRao hospitalinDelhi,IndiawasdeterminedusingRocheAmplicorMultiWellPlatekit(MWP)aswellasusingin-house PCRassay.Weused593endocervicalswabsforclinicalevaluationofthein-housedevelopedassayagainstDirect FluorescenceAssay(DFA;GroupIn=274)andRocheAmplicorMWPkit(GroupII,n=319samples)and determinedthesensitivity,specificity,positivepredictivevalue(PPV),negativepredictivevalue(NPV)ofthein- housedevelopedassay. Results:Wedetected23.0%positivecasesandtherewasahigherrepresentationofwomenaged18-33inthis group.Anin-housePCRassaywasdevelopedandevaluatedbytargetinguniquesequencewithinthegyrAgene ofC.trachomatis.SpecificityofthereactionwasconfirmedbyusinggenomicDNAofhumanandotherSTIrelated microorganismsastemplate.Assayishighlysensitiveandcandetectaslowas10fgofC.trachomatisDNA.The resolvedsensitivityofin-housePCRwas94.5%comparedwith88.0%ofDFAassay.Thehighspecificity(98.4%)and sensitivity(97.1%)ofthein-houseassayagainstRochekitandavailabilityoftestresultswithin3hoursallowedfor immediatetreatmentandreducedtheriskofpotentialonwardtransmission. Conclusions:Thein-housePCRmethodiscosteffective(~20.0%ofRocheassay)andhencecouldbeabetter alternativeforro
