Rif1SupportstheFunctionoftheCSTComplexinYeast TelomereCapping SavaniAnbalagan,DiegoBonetti,GiovannaLucchini,MariaPiaLonghese* DipartimentodiBiotecnologieeBioscienze,Universita` diMilano-Bicocca,Milano,Italy Abstract TelomereintegrityinbuddingyeastdependsontheCST(Cdc13-Stn1-Ten1)andshelterin-like(Rap1-Rif1-Rif2)complexes, whicharethoughttoactindependentlyfromeachother.Hereweshowthataspecificfunctionalinteractionindeedexists amongcomponentsofthetwocomplexes.Inparticular,unlikeRIF2deletion,thelackofRif1islethalforstn1DCcellsand causesadramaticreductioninviabilityofcdc13-1andcdc13-5mutants.ThissyntheticinteractionbetweenRif1andtheCST complexoccursindependentlyofrif1D-inducedalterationsintelomerelength.Bothcdc13-1rif1Dandcdc13-5rif1Dcells displayveryhighamountsoftelomericsingle-strandedDNAandDNAdamagecheckpointactivation,indicatingthatsevere defectsintelomereintegritycausetheirlossofviability.Inagreementwiththishypothesis,bothDNAdamagecheckpoint activationandlethalityincdc13rif1DcellsarepartiallycounteractedbythelackoftheExo1nuclease,whichisinvolvedin telomericsingle-strandedDNAgeneration.ThefunctionalinteractionbetweenRif1andtheCSTcomplexisspecific,because RIF1deletiondoesnotenhancecheckpointactivationincaseofCST-independenttelomerecappingdeficiencies,suchas thosecausedbytheabsenceofYkuortelomerase.Thus,thesedatahighlightanovelroleforRif1inassistingtheessential telomereprotectionfunctionoftheCSTcomplex.
