Bramham* DepartmentofBiomedicineandBergenMentalHealthResearchCenter,UniversityofBergen,Bergen,Norway Abstract Progenitorcellsintheadultdentategyrusprovideaconstantsupplyofneuronalprecursors,yetonlyasmallfractionof thesecellssurviveanddevelopintomaturedentategranulecells(DGCs).Amajorchallengeofcurrentresearchisthusto understandthestringentselectionprocessthatgovernsthematurationandfunctionalintegrationofadult-bornDGCs.In matureDGCs,high-frequencystimulation(HFS)oftheperforantpathinputelicitsrobustexpressionoftheimmediateearly geneArc/Arg3.1,traffickingofitsmRNAtodendrites,andlocalsynthesisoftheproteinnecessaryforconsolidationoflong- term potentiation (LTP). Given the synaptic commitment inherent in LTP consolidation, we considered that HFS-evoked expressionofArccouldbeusedtotimemapthefunctionalintegrationofnewbornDGCs.Dividingcellswerebirthmarked by BrdU-labeling at 1, 7, 14, 21, or 28days prior to induction of LTP and expression of Arc was examined by confocal microscopy.Contrarytoexpectation,LTPdidnotinduceArcexpressioninnewborncellsatanyage,suggestingtheymight berefractorytosynaptically-evokedArcexpressionforatleastonemonth.Importantly,however,spontaneousexpression ofArcwasdetectedinBrdU-labeledcellsandstronglyassociatedwiththesurvivalandmaturationofNeuN-positiveDGCs. Moreover,Arcexpressionattheearliestages(1and7days),clearlyprecedestheformationofglutamatergicsynapseson newneurons.TheseresultssuggestanunexpectedearlyroleforArcinadult-bornDGCs,distinctfromitsfunctionsinLTP, LTD,andhomeostaticsynapticplasticity.
