Kienesbergeretal.GutPathogens2012,4:9 /content/4/1/9 RESEARCH OpenAccess SerologichostresponsetoHelicobacterpyloriand CampylobacterjejuniinsociallyhousedRhesus macaques(Macacamulatta) SabineKienesberger1*,GuillermoIPerez-Perez1,2,JuanLRivera-Correa5,RafaelTosado-Acevedo6,HuilinLi4, AndreDubois7,JanisAGonzalez-Martinez8,MariaGloriaDominguez-Bello1,5andMartinJBlaser1,2,3 Abstract Background:Helicobacterpyloriaresuccessfulcolonizersofthehumangastricmucosa.Colonizationincreasesthe riskofpepticulcerdiseaseandadenocarcinoma.However,potentialbenefitsofH.pyloricolonizationinclude protectionagainstearly-onsetasthmaandagainstgastrointestinalinfections.Campylobacterjejuniarealeading causeofbacterialdiarrheaandcomplicationsincludeGuillain-Barrésyndrome.Here,wedescribethedevelopment ofreliableserologicalassaystodetectantibodiesagainstthosetwobacteriainRhesusmacaquesandinvestigated theirdistributionwithinasocialgroupofmonkeys. Methods:Twocohortsofmonkeyswereanalyzed.Thefirstcohortconsistedof30monkeysandwasusedto establishanenzyme-linkedimmunosorbentassay(ELISA)forH.pyloriantibodiesdetection.Toevaluatecolonization ofthosemacaques,stomachbiopsieswerecollectedandanalyzedforthepresenceofH.pyloribyhistologyand culture.C.jejuniELISAswereestablishedusinghumanserumwithknownC.jejuniantibodystatus.Next,plasma samplesofthe89macaques(Cohort2)wereassayedforantibodiesandthenstatisticallyanalyzed. Results:AnH.pyloriIgGELISA,whichwas100%specificand93%sensitive,wasestablished.Incontrast,theIgAELISA wasonly82%specificand61%sensitive.TheCagAIgGassaywas100%sensitiveand61%ofthemacaqueswere positive.Incohort2,62%macaqueswereH.pylorisero-positiveand52%wereCagApositive.TheprevalenceofH.pylori IgGandCagAIgGincreasedwithmonkeyageasdescribedforhumans.Ofthe89macaques52%showedIgGagainstC. jejunibutincontrasttoH.pylori,thesero-prevalencewasnotassociatedwithincreasingage.However,therewasadrop intheIgG(butnotinIgA)meanvaluesbetweeninfantandjuvenilemacaques,similartotrendsdescribedinhumans.
