Abstract
Background:Theamygdala,hippocampus,medialprefrontalcortex(mPFC)andbrain-stemsubregionsare
implicatedinfearconditioningandextinction,andarebrainregionsknowntobesexuallydimorphic.Weused
functionalmagneticresonanceimaging(fMRI)toinvestigatesexdifferencesinbrainactivityintheseregionsduring
fearconditioningandextinction.
Methods:Subjectswere12healthymencomparableto12healthywomenwhounderwenta2-dayexperimentin
a3TMRscanner.Fearconditioningandextinctionlearningoccurredonday1andextinctionrecalloccurredon
day2.Theconditionedstimuliwerevisualcuesandtheunconditionedstimuluswasamildelectricshock.Skin
conductanceresponses(SCR)wererecordedthroughouttheexperimentasanindexoftheconditionedresponse.
fMRIdata(blood-oxygen-level-dependent[BOLD]signalchanges)wereanalyzedusingSPM8.
Results:FindingsshowednosignificantsexdifferencesinSCRduringanyexperimentalphases.However,during
fearconditioning,thereweresignificantlygreaterBOLD-signalchangesintherightamygdala,rightrostralanterior
cingulate(rACC)anddorsalanteriorcingulatecortex(dACC)inwomencomparedwithmen.Incontrast,men
showedsignificantlygreatersignalchangesinbilateralrACCduringextinctionrecall.
Conclusions:Theseresultsindicatesexdifferencesinbrainactivationwithinthefearcircuitryofhealthysubjects
despitesimilarperipheralautonomicresponses.Furthermore,wefoundthatregionswheresexdifferenceswere
previouslyreportedinresponsetostress,alsoexhibitedsexdifferencesduringfearconditioningandextinction.
