ShrimpAlpha-2-MacroglobulinPreventstheBacterial EscapebyInhibitingFibrinolysisofBloodClots VorraponChaikeeratisak,KunlayaSomboonwiwat,AnchaleeTassanakajon* DepartmentofBiochemistry,FacultyofScience,CenterofExcellenceforMolecularBiologyandGenomicsofShrimp,ChulalongkornUniversity,Bangkok,Thailand Abstract ProteomicanalysisofthehemocyticproteinsofPenaeusmonodon(Pm)haspreviouslyshownthatalpha-2-macroglobulin (A2M)wasamongtheproteinsthatshowedsubstantiallyalteredexpressionlevelsuponVibrioharveyiinfection.Therefore, inthisstudyitspotentiallyimportantroleintheresponseofshrimptobacterialinfectionwasfurthercharacterized.The yeasttwo-hybridsystemrevealedthatthereceptorbindingdomainofPmA2Minteractedwiththecarboxyl-terminusofone orbothofthetransglutaminasetypeIIisoforms,whicharekeyenzymesinvolvedintheshrimpclottingsystem.Inaccord withthis,PmA2Mwasfoundtobelocalizedontheextracellularbloodclotsandtocolocalizewithclottableproteins.RNA interference (RNAi)-mediated knockdown of A2M transcript levels reduced the PmA2M transcript levels (,94%) and significantlyreducedthebacterialseizingabilityoftheclottingsystem,resultinginanupto3.3-foldhighernumberofV. harveyithatsystemicallydisseminatedintothecirculatorysystemat5minpost-infectionbeforesubsequentclearanceby the immune system. Furthermore, an appearance of PmA2M depleted clots in the presence of V. harveyi strikingly demonstratedfibrinolysiszonessurroundingthebacteria.ThisstudyprovidesthefirstevidenceofthevitalroleofPmA2M inenhancingbacterialsequestrationbyprotectingbloodclotsagainstfibrinolysis.
