Abstract Introduction:TNF-likeweakinducerofapoptosis(TWEAK)hasbeenproposedasamediatorofinflammationand boneerosioninrheumatoidarthritis(RA).ThisstudyaimedtoinvestigateTWEAKandTWEAKreceptor(Fn14) expressioninsynovialtissuefrompatientswithactiveandinactiverheumatoidarthritis(RA),osteoarthritis(OA)and normalcontrolsandassesssoluble(s)TWEAKlevelsinthesynovialfluidsfrompatientswithactiveRAandOA. EffectsofsTWEAKonosteoclastsandosteoblastswereinvestigatedinvitro. Methods:TWEAKandFn14expressionweredetectedinsynovialtissuesbyimmunohistochemistry(IHC).Selected tissueswereduallabelledwithantibodiesspecificforTWEAKandlineage-selectivecellsurfacemarkersCD68, TryptaseG,CD22andCD38.TWEAKmRNAexpressionwasexaminedinhumanperipheralbloodmononuclearcells (PBMC)sortedonthebasisoftheirexpressionofCD22.sTWEAKwasdetectedinsynovialfluidfromOAandRA patientsbyELISA.TheeffectofsTWEAKonPBMCandRAW264.7osteoclastogenesiswasexamined.Theeffectof sTWEAKoncellsurfacereceptoractivatorofNFKappaBLigand(RANKL)expressionbyhumanosteoblastswas determinedbyflowcytometry. Results:TWEAKandFn14expressionweresignificantlyhigherinsynovialtissuefromallpatientgroupscompared tothesynovialtissuefromcontrolsubjects(P0.05).TWEAKwassignificantlyhigherinactivecomparedwith inactiveRAtissues(P0.05).TWEAKexpressionco-localisedwithasubsetofCD38+ plasmacellsandwithCD22+ B-lymphocytesinRAtissues.AbundantTWEAKmRNAexpressionwasdetectedinnormalhumanCD22+ Bcells.
