青海大学硕士学位论文 Abstract Abstract Objective: By measuring supernatant of lung tissue interleukin-6(IL-6), inter- leukin-8(IL-8) and interleaukin-10(IL-10) level of Wistar rats at different altitudes in the same time period, to examine the effects of airway inflammation and tissue injury. Methods ：40 Wistar rats were bought from College of Gansu Traditional Chinese Me- dicine, all these rats were randomly divided into 3 Group: Comparison Group(n=8, Lanzhou,1500m),Medium Altitude Group(n= 16,Xining,2260m), Simulation High Alt- itude Group(n= 16,Low pressure oxygen tank,5000m). The Comparison Group’s lung tissue were taken in Lanzhou. Medium Altitude Group was fed in QingHai University Medical School animal room at 2260m altitude, by day and night, light and airy, the room temperature is about 25 ℃, regularly disinfected, the rats were free feeding and drinking water. Simulation High Altitude Group was raised in low pressure oxygen tank that at high altitude Qinghai University Medical Center School of Medicine, the low pressure oxygen tank condition is: simulated 5000m alt- itude, pressure: 397.5mmHg, oxygen partial pressure: 52889Pa～53001Pa, tempera- ture: 16.9℃-24 ℃, humidity: 37.3%-51%. Animal’s feeding, drinking, cleaning and disinfection were carried out in the low pressure oxygen tank. The rats of Medium Group and Simulation High Altitude Group were divided into 4 groups by the day we observed: 1 day, 30 days, every group have 8 rats, This research project will collected the supernatant lung tissue homogenate of the 1 day and 30 days. By using Enzyme-Linked Immuno Sorbent Assay (ELISA) to test the levels of the IL-6,IL-8 and IL-10.
