HuanWang1,MeishanZhang2,RuiGuo3,DechengShi1,BaoLiu1,XiuyunLin4andChunwuYang1* Abstract Background:Itiswellknownthatsaltstresshasdifferenteffectsonoldandyoungtissues.However,itremains largelyunexploredwhetheroldandyoungtissueshavedifferentregulatorymechanismduringadaptationofplants tosaltstress.Theaimofthisstudywastoinvestigatewhethersaltstresshasdifferenteffectsontheionbalance andnitrogenmetabolismintheoldandyoungleavesofrice,andtocomparefunctionsofbothorgansinricesalt tolerance. Results:RiceprotectedyoungleavesfromionharmviathelargeaccumulationofNa+andCl-inoldleaves.The up-regulationofOsHKT1;1,OsHAK10andOsHAK16mightcontributetoaccumulationofNa+inoldleavesundersalt stress.Inaddition,lowerexpressionofOsHKT1;5andOsSOS1inoldleavesmaydecreasefrequencyofretrievingNa+ fromoldleafcells.Undersaltstress,oldleavesshowedhigherconcentrationofNO3- contentthanyoungleaves. Up-regulationofOsNRT1;2,agenecodingnitratetransporter,mightcontributetotheaccumulationofNO3- inthe oldleavesofsaltstressed-rice.Saltstressclearlyup-regulatedtheexpressionofOsGDH2andOsGDH3inoldleaves, whilestronglydown-regulatedexpressionofOsGS2andOsFd-GOGATinoldleaves. Conclusions:Thedown-regulationofOsGS2andOsFd-GOGATinoldleavesmightbeaharmfulresponseto excessesofNa+andCl-.Undersaltstress,ricemightaccumulateNa+andCl-totoxiclevelsinoldleaves.Thismight + influencephotorespirationprocess,reduceNH productionfromphotorespiration,andimmediatelydown-regulate 4 theexpressionofOsGS2andOsFd-GOGATinoldleavesofsaltstressedrice.ExcessesofNa+andCl-alsomight + changethepathwayofNH assimilationinoldleavesofsaltstressedriceplants,weakenGOGAT/GSpathwayand 4 elevateGDHpathway.
