Results:Weinvestigatedtheresponsesofthedrought-sensitivepotatocultivarWhiteLadyandthedrought- tolerantTPS1transgenicvarianttoprolongeddroughtstressatboththetranscriptionalandmetaboliclevels.Leaf mRNAexpressionprofileswerecomparedusingthePOCImicroarray,whichcontains42,034potatounigene probes.Weidentified379genesofknownfunctionthatshowedatleasta2-foldchangeinexpressionacross genotypes,stresslevelsortheinteractionbetweenthesefactors.Wild-typeleaveshadtwiceasmanygeneswith alteredexpressioninresponsetostressthanTPS1transgenicleaves,but112genesweredifferentiallyexpressedin bothstrains.Weidentified42transcriptionfactorgeneswithalteredexpression,ofwhichfourwereuniquelyup- regulatedinTPS1transgenicleaves.Themajorityofthegeneswithalteredexpressionthathavebeenimplicatedin photosynthesisandcarbohydratemetabolismweredown-regulatedinboththewild-typeandTPS1transgenic plants.Inagreementwiththisfinding,thestarchconcentrationofthestressedleaveswasverylow.Atthe metaboliclevel,thecontentsoffructose,galactoseandglucosewereincreasedanddecreasedinthewild-typeand TPS1transgenicleaves,respectively,whiletheamountsofproline,inositolandraffinosewerehighlyincreasedin boththewild-typeandTPS1transgenicleavesunderdroughtconditions. Conclusions:Toourknowledge,thisstudyisthemostextensivetranscriptionalandmetabolicanalysisofa transgenic,drought-tolerantpotatoline.Weidentifiedfourgenesthatwerepre
