Methods:TheexpressionofMK-1andRegⅣwasdetectedbyimmunohistochemicalmethodinparaffin- embeddedsectionsofsurgicalresectedspecimensfromgallbladderadenocarcinoma(n=108),peritumoraltissues (n=46),adenomatouspolyp(n=15),andchroniccholecystitis(n=35). Results:ThepositiverateofMK-1orRegⅣexpressionwassignificantlyhigheringallbladderadenocarcinoma thanthatinperitumoraltissues(c2MK-1 =18.76,P0.01;c2RegⅣ =9.92,P0.01),denomatouspolyp(c2MK-1 = 9.49,P0.01;c2RegⅣ =8.59,P0.01)andchroniccholecystitis(c2MK-1 =24.11,P0.01;c2RegⅣ =19.24,P0.01). ThepositivecasesofMK-1and/orRegⅣinthebenignlesionsshowedmoderately-orsevere-atypicalhyperplasia ofgallbladderepitheli.ThepositiveratesofMK-1weresignificantlyhigherinthecasesofwell-differentiated adenocarcinoma,no-metastasisoflymphnode,andno-invasivenessofregionaltissuesthanthoseintheonesof differentiatedadenocarcinoma,metastasisoflymphnode,andinvasivenessofregionaltissuesingallbladder adenocarcinoma(P0.05orP0.01).Onthecontrary,thepositiveratesofRegⅣweresignificantlylowerinthe casesofwell-differentiatedadenocarcinoma,no-metastasisoflymphnode,andno-invasivenessofregionaltissues thanthoseintheonesofdifferentiatedadenocarcinoma,metastasisoflymphnode,andinvasivenessofregional tissuesingallbladderadenocarcinoma(P0.05orP0.01).UnivariateKaplan-Meieranalysisshowedthat decreasedexpressionofMK-1(P=0.09)orincreasedexpressionofRegⅣ(P=0.003)wasassociatedwith decreasedoverallsurvival.MultivariateCoxregressionanalysisshowedthatdecreasedexpressionofMK-1(P= 0.033)andincreasedexpressionofRegⅣ(P=0.008)wasanindependentprognosticpredictoringallbladder adenocarcinoma.
