原发胃癌中p15、p16基因蛋白的表达及临床意义 　　作者：高云升 作者单位：盖州市中心医院普通外科,辽宁 盖州 115200 　　【摘要】 目的 检测原发胃癌原发灶及癌旁正常组织中p15、p16基因蛋白表达情况,探讨p15、p16基因失活与胃癌发生发展及临床病理特征的关系。方法 应用免疫组织化学方法检测32例进展期胃癌石蜡标本中p15、p16 的表达，并探讨其表达与临床病理特征之间的关系。结果 p16、p15 在原发灶与癌旁正常组织中阳性表达率差异有统计学意义(P0.05)，p15、p16 基因表达在有无淋巴结转移和不同TNM分期的病例之间差异均有统计学意义(P0.05)。结论 p15、p16 蛋白表达可能与胃癌癌变过程相关,并可以作为临床治疗提供理论依据。 　　【关键词】 胃肿瘤 免疫组织化学 p15 p16 　　The Clinical Significance and Expression of p15 as 　　Well as p16 in Primary Gastric CancerGAO Yun-sheng 　　(Department of General Surgery, Gaizhou Central Hospital, Gaizhou 115200 China)【Abstract】 Objective To test the expression of p15 and p16 in primary gastric cancer and its normal tissue and to discuss the relation between the genetic losing activity of p15 together with p16 and the development of gastric cancer as well as the features of clinical pathology. Methods 32 cases of the expression of p15 and p16 in samples of paraffin wax of gastric cancer were tested by immunohistochemistry method and tried to discuss the relationship between the expression of p15 and p16 and the features of clinical pathology. Results It had the significance in terms of statistics in the difference in positive expression rates of p15 and p16 in primary gastric carcinoma and normal tissue of adjacent to cancer (P0.05) as well as the difference between the cases with lymphnode metastasis or not and different clinical pathological stages (P0.05). Conclusions The protein expression of p15 and p16 may be related to the cancerization of gastric cancer, and it can be used for providing the theoretical evidence as the clinical treatment. 　　【Key words】 gastric carcinoma;immunohistochemistry;p15; p16 　　国内外研究提示,胃癌的发生发展是一个多基因共同参与的多突变的过程。但具体的癌变机制目前尚不完全明确[1]。p16、p15 定位于人第九号染色体21区域。其为细胞周期的负性调节因子,阻止细胞进入S期,故可能在肿瘤的发生发展中起重要作用[2]。本研究采用免疫组织化学方法,检测32例原发胃癌原发灶与癌旁正常组织中胃癌p15、p16基因蛋白表达情况，旨在探讨p15、p16基因失活与胃癌发生发展及与临床病理特征的关系，为胃癌的治疗提供理论依据。 　　1 材料与方法 　　1.1 临床资料 　　收集盖州市中心医院2005年8月至2007年5月收治并行手术治疗的32例胃癌患者胃癌组织及癌旁正常组织标本。其
