波长切换高效液相色谱法同时测定壮骨关节丸中7种有效成分的含量 颜冬梅1 王跃飞1 刘亚男1 谭沛2 潘桂湘1 代志2 高秀梅1* （1天津中医药大学中医药研究院，现代中药发现与制剂技术教育部工程研究中心，天津 300193； 2 三九医药股份有限公司，深圳，518029） 摘 要 目的：采用波长切换高效液相色谱法，建立同时测定壮骨关节丸中7种有效成分含量的方法。方法：色谱柱：Zorbax SB-C18（4.6×250 mm, 5 μm）；流动相：0.05%甲酸水溶液-乙腈梯度洗脱；检测波长：0~25 min：283 nm；25.01~30 min：365 nm；30.01~32 min：270 nm；32.01~37 min：246 nm；37.01~65 min：322 nm；柱温：40 ℃；流速：1.0 mL/min。结果：柚皮苷、槲皮素、淫羊藿苷、补骨脂素、异补骨脂素、芒柄花素和蛇床子素在相应的浓度范围内与峰面积呈良好线性关系，相关系数均大于0.9997；平均回收率（n=6）在97.77%~ 104.3%相对标准偏差（RSD）均小于4.01%。结论：本方法简便快速、准确、精密度高、重复性好，适用于壮骨关节丸的质量控制。 关键词 波长切换；高效液相色谱法；壮骨关节丸 Simultaneous determination of seven active compounds in Zhuanggu-Guanjie pill by HPLC using switching UV wavelength (1 The TCM Research Centre of Tianjin University of TCM, Engineering Research Center of Modern Chinese Medicine Discovery and Preparation Technique，Ministry of Education, Tianjin 300193, China; 2 Sanjiu Medical Pharmaceutical CO., LTD., Shenzhen 518029, China) YAN Dong-Mei1, WANG Yue-Fei1, LIU Ya-Nan1, TAN Pei2, PAN Gui-Xiang1, DAI Zhi2, GAO Xiu-Mei1* Abstract Objective: To establish a method for simultaneous determination of seven active components in Zhuanggu-Guanjie pill using switching UV wavelength. Method: The HPLC analysis performed on Zorbax SB-C18 column by using the mobile phase composed of acetonitrile and 0.05% formic acid water in gradient elution. Detection wavelength was set as follow: 283nm at 0~25min, 365nm at 25.01~30min, 270nm at 30.01~32min, 246nm at 32.01~37min, 322nm at 37.01~65min. The column temperature was maintained at 40°С. The flow rate was fixed at 1.0 mL/min. Results: Naringin, quercetin, icariin, psoralen, isopsoralen, biochanin and osthole showed good linearity over the tested ranges (r≥0.9997). The average recoveries were ranged from 97.77 to 104.3% with RSDs ≤4.01%. Conclusion: This method was qualified for analysis of Zhuanggu-Guanjie pill, which had advantage of accuracy, repeatability and simplicity.
