The expression and significance of VEGF-A in Serum and tumor tissue of nude mouse with colorectal transplantation tumor ABSTRACT Objective: to explore the expression of VEGF-A in the serum and tumor tissue of colorectal cancer SW480 cell lines nude mouse transplantation tumor, and observe whether it is associated with the occurrence of colorectal cancer development, analyse the correlation of nude mice serum and transplanted tumor tissue on VEGF-A, and to determine whether the tissue VEGF-A measurement can be replaced by serum VEGF-A measurement, in order to provide new experimental and theoretical basis to the the determination of treatment and prognosis of colorectal cancer. Methods: 20 nude mice of SPF were randomly divided into two groups: model group and healthy group, each group has 10 mice. Human colorectal cancer SW480 cell lines nude mouse transplantation tumor model is set up in model group, observe the emergence and growth of transplanted tumor. Get 2ml blood sample from aorta abdominalis of the two groups after transplanted tumor grow successfully, and remove the subcutaneous tumor of the model group, and take the normal colon tissue of healthy group as contrast, and to detect the expression of VEGF-A in the serum and tumor tissue by ELISA method and immunohistochemical SP method. Results: the tumor formation rate in this experiment is 100%, tumors had an average of 5 mm diameter. The serum VEGF-A level of model mice group(426.26±160.91pg/ml)is significantly higher than the healthy group(139.11±44.45pg/ml), differences are statistically significant(t=5.439 ， P0.001); the positive expression rate of VEGF-A in tumor
