园 艺 学 报 2012 ，39 （3 ）：588 –592 http: // www. ahs. ac. cn Acta Horticulturae Sinica E-mail: yuanyixuebao@126.com 亚洲百合与青岛百合杂交及其 FISH 快速鉴定 任贵玲，张线线，周树军* （浙江大学农业与生物技术学院园艺系，农业部园艺植物生长发育与品质调控重点开放实验室，杭州 310058 ） 摘 要：利用切柱头授粉和胚抢救技术获得亚洲百合‘Petit Brigitte ’× 青岛百合的 14 个杂交后代， 以 45S rDNA 为探针对亲本和其中 4 个后代进行了荧光原位杂交（FISH ）分析，结果表明，亲本和后代 都为二倍体（2n = 2x = 24 ）；‘Petit Brigitte ’有 10 个 45S rDNA 位点，分别位于 1、2 、4 、5 和 7 号染色 体上；青岛百合有 8 个 45S rDNA 位点，分别位于 3、4 、5 和 10 号染色体上；4 个杂种后代有 9 个 45S rDNA 位点，分别源于‘Petit Brigitte ’和青岛百合。 关键词：百合；远缘杂交；rDNA ；荧光原位杂交 中图分类号：S 682.2 文献标识码：A 文章编号：0513-353X （2012 ）03-0588-05 Hybridization and Identification Between Asiatic Lily and Lilium tsingtauense Using FISH * REN Gui-ling ，ZHANG Xian-xian ，and ZHOU Shu-jun （The Key Laboratory of Horticultural Plant Growth ，Development and Quality Improvement ，the Ministry of Agriculture of the People’s Republic of China ；Department of Horticulture ，College of Agriculture and Biotechnology ，Zhejiang University，Hangzhou 310058 ，China ） Abstract ：Hybridization between Asiatic lily ‘Petit Brigitte ’× Lilium tsingtauense was conducted with cut style pollination and embryo rescue. Fourteen progenies were obtained，and the parents and 4 progenies were analyzed using FISH with 45S rDNA as probe. The results showed that all of them are diploid （2n = 2x = 24 ）；‘Petit Brigitte ’has 10 loci of 45S rDNA distributed on chromosome CH-1 ，CH-2，CH-5 and CH-7 and Lilium tsingtauense has 8 on CH-3 ，CH-4，CH-5 and CH-10 respectively，2n = 2x = 24 for Lilium tsingtauense and 2n = 2x = 24 for hybrids ，respectively ；all four progenies have 9 loci of 45S rDNA ，unambiguously ，coming from ‘Petit Brigitte ’and Lilium tsingtauense respectively.
