The characteristics of agarose gel
Natural AGAR AGAR () is a kind of polysaccharide, mainly by agarose (agarose, accounts for about 80%) and AGAR gel (agaropectin). Agarose was composed of galactose and its derivatives neutral material, without charge, and AGAR gel is a kind of strongly acidic polysaccharides containing sulfate and carboxyl, because these groups with charge, under the effect of electric field can produce strong electroosmosis phenomenon, combined with sulfuric acid root with certain proteins effect and influence of electrophoresis rate and separation effect. Therefore, the current use of agarose to electrophoresis support for flat electrophoresis, its advantages are as follows.
(1) agarose gel electrophoresis is simple, the electrophoresis is fast, and the sample can be electrophoresis without preprocessing.
(2) the agarose gel evenly, water content (about 98% ~ 98%), approximate free electrophoresis, sample diffusion is free current, a mere adsorption of the sample, so the electrophoresis is clear, high resolution, good repeatability.
(3) agarose is transparent and free of ultraviolet absorption, electrophoresis process and result can be detected by ultraviolet light and quantitative determination.
(4) after electrophoresis, the zone is easy to stain, and the sample is easy to be cleaned, which is convenient for quantitative determination. The dry film can be preserved for a long time.
At present, agarose is commonly used as electrophoretic support to separate proteins and isoenzymes. Combining the agarose electrophoresis and immunochemical, develop immunoelectrophoresis technique, can identify other methods cant identify complex system, the setting up of ultramicro technology, 0.1 ug protein can be checked out.
