提 要 目的：探讨纳米雄黄对人皮肤鳞癌A431 细胞株增殖抑制及诱导凋亡的作用，进 一步阐明纳米雄黄诱导肿瘤细胞凋亡的机理。 方法：应用MTT 比色法、细胞形态学观察、流式细胞仪、RT-PCR 技术观察纳 米雄黄体对人皮肤鳞状细胞癌 A431 细胞的增殖抑制、诱导凋亡作用。 结果：纳米雄黄具有诱导皮肤鳞癌A431 细胞凋亡和抑制细胞增殖作用，且随着 纳米雄黄浓度的增高作用增强；纳米雄黄与顺铂联合应用对诱导 A431 细胞凋亡、抑 制增殖有协同作用。 结论：纳米雄黄可通过诱导肿瘤细胞凋亡和抑制其增殖发挥抗肿瘤作用；纳米雄 黄与顺铂联合应用可能产生协同作用；纳米雄黄诱导肿瘤细胞凋亡的作用机理可能与 上调 Caspase-3 的表达和下调 Survivin 的表达有关。 关键词 纳米雄黄；人皮肤鳞状细胞癌 A431 细胞株；诱导凋亡；增殖抑制 Experimental study of proliferation inhibition and apoptosis induced by realgar nanoparticles on skin squamous cell carcinoma A431 cells Speciality：Clinical internal Medicine of TCM Western Medicine Author ：Nie Ben Tutor：Professor Qi Yuanfu Abstract Objective: the study observes inhibiting proliferation and inducting apoptosis effect of nanometer realgar on human skin squamous cell carcinoma A431 cell, and explore the mechanism of nanometer realgar inducing the human skin squamous cell carcinoma A431 apoptosis.
