心肌联合注射纤维蛋白封闭剂在细胞移植中应用研究.docVIP

【摘要】 　　目的：探讨心肌注射骨髓间充质干细胞（mesenchymal stem cells ，M SCs ）联合应用纤维蛋白封闭剂（FibrinSealant，FS ），对细胞滞留量的影响。方法：（1 ）提取、分离、培养、扩增小型猪骨髓M SCs 备用；（2 ） 小型猪随机分为2 组，成功建立急性心肌梗塞模型，然后分为单纯M SCs 移植组和M SCs 联合纤维蛋白封闭剂移植组，对比移植6 周后细胞的滞留量及新生血管的密度，超声学评价心脏功能。结果：6 周后M SCs +FS 组荧光标记的M SCs数量、新生血管密度均较M SCs 组显著增多（ P ＜ 0101 ），M SCs +FS 组左室短轴缩短率（FS %）、心肌射血分数（EF %）较M SCs 组显著提高（P ＜ 0105 ，＜ 0101 ）。结论：经心外膜心肌注射M SCs 联合纤维蛋白封闭剂较单纯心肌注射M SCs 能够明显提高移植细胞的滞留存活数量，增加新生血管密度，改善心肌收缩功能。 【关键词】 心肌梗塞；干细胞移植；心室功能 　　Experimental studies of intramyocardial injecting bone marrow mesenchymal stem cells combined with fibrin sealant in cells transplantation 　　Z H A O Jian ， W A N G Ying ， W A N G Guang2li，C H E Hui，K O N G Xiang2rong ，LIU Xiao2cheng Chinese Journal of Cardiovascular Rehabilitation M edicine ，2007 ，16 （4 ）：321 Abstract：Objective ：To investigate the effects ofintramyocardialinjecting bone marrow mesenchy mal stem cells com2 bined with fibrin sealant in cells transplantation .M ethods ：（1 ）Prepared for isolation ，purification ，culture ，expan2 sion of bone marrow M SCs1 （2 ）Mini2swine were randomly assigned to establish models of acute myocardialinfarction and then were performed by the subsequent procedure ：intramyocardial M SCs injection alone（Group M SCs ）and in2 tramyocardial M SCs injection com bined with FS （Group M SCs +FS ）.Six weeks postoperatively ，quantitative analysis of fluorescent2labeled cells was performed by Image Pro Plus 415 software .Likewisely ，neovascularization by histo2 chemically staining was analyzed as well.Cardiac function was assessed by echocardiorgaphy .Results ：Six weeks postoperatively ，both the retention nu m ber of positively labeled cells and new vessels density in Group M SCs +FS showed significant increase com pared with those in Group M SCs （P ＜ 0101 ）.M yocardial function of EF % and FS % in Group M SCs +FS markedly im proved co m pared with that in Group M SCs （ P ＜ 0105 ，＜ 0101 ）.Conclusion ：In2 tramyocardialinjecting bone marrow M CSs co m bined with fibrin sealant
