生物技术通报 ·研究报告 · B IO TECHN OLO G Y　BULL E T IN 2009年第 10 期 基于氨单加氧酶基因的自养脱氮菌群结构分析 郑雪松 龚钢明 (上海应用技术学院 ,上海 200235) 　　摘 　要 : 　全程 自养脱氮是一种在高氨氮低溶氧条件下完全由自养菌群作用脱除氮素的现象 。以全程 自养脱氮污泥为 研究对象 ,特异性扩增氨单加氧酶活性基因 am oA 片段 ,建立克隆文库并对克隆序列进行系统发育学分析 ,考察全程 自养脱氮 系统从建立到退化过程中氨氧化菌的结构变迁 。结果表明: N itrosom onas olig otrop ha 和 N itrosom onas eu rop aea 细菌是系统中的 主要氨氧化菌 ,而随着系统的退化前者逐渐被后者完全取代 ,而氨氧化菌的种群变迁可能并不是全混流系统全程 自养脱氮效 率下降的原因。 关键词 : 　氨氧化菌 氨单加氧酶基因 自养脱氮 克隆文库 Phylogenetic Ana lysis ba sed on the am oA Gene of Amm on ia O x id izers in an Autotroph ic N itrogenRem ova l Reactor Zheng Xue song Gong Gangm ing (S hangha i Institu te of Technology, S hangha i 200235) 　　Ab s tra c t: 　 In order to analyze ammon iaoxidizing pop u lation in deammon ificationsludge, a p artial stretch of the gene encoding the activesite polyp ep tide of ammon ia monooxygena se ( am oA ) wa s amp lified and the gene lib rarie s were con structed The re su lt of gene sequences and phylogenetic analysis showed that two sp ecie s of ammon iaoxidizing bacteria, including N itrosom onas olig otrop ha and N itrosom onas eu rop aea are the dom inant sp ecie s in the deammon ification reactor.
