EFFECTS OF SEVOFLURANE ON P65, CD8 RECEPTOR AND LUNG FUNCTION IN LUNG TISSUE OF CHRONIC OBSTRUCTIVE PULMONARY DISEASE RAT ABSTRACT Objective: To explore the influence of sevoflurane on P65, CD8 receptor and lung function in lung tissue of COPD model rats and to discuss its effects on lung protection and related mechanisms of molecular action. Methods: Thirty two male SD rats were randomly divided into control group, COPD group, 1% sevoflurane COPD group and 2.5% sevoflurane COPD group respectively, 8 rats each. After establishing COPD model by passive smoking and tracheal instillation of LPS, rats in SEVOCOPD groups were put into anesthetic induction container connected by closed tubes and inhaled 1% or 2.5% sevoflurane for 1 hour respectively. All rats underwent lung function test by lung compliance analyzer 1.0. Their slides of lung tissue were fixed and their inflammatory pathological changes were inspected under transmission electron microscope. The expression of P65 and CD8 receptor was measured by immunohistochemistry. Furthermore, P65 was measured by ECL-Western-blot method respectively. They all received semi-quantity analysis by applying Image-Pro-plus 6.0 software. The differences of rats’ lung function, CD8 and P65 in all groups underwent statistic analysis. Results: Type II epithelial cells in SEVOCOPD group proliferated more significantly than those in COPD group while the degree of macrophage’s aggregation in SEVOCOPD group was less significant.
