BRCA1在乳腺肿瘤中表达及其临床意义 　 作者：黄耀，魏杨辉，赵洁，李建文 【摘要】 目的 探讨乳腺癌易感基因(BRCA1）在乳腺癌和乳腺良性肿瘤中的表达及其临床意义。方法 应用ABC免疫组化法检测52例乳腺癌、30例乳腺纤维腺瘤并上皮增生活跃和10例乳腺增生症患者中的BRCA1的表达。结果 BRCA1在乳腺癌中的阳性表达率为38%（20/52），在乳腺纤维腺瘤中的阳性表达率为73%（22/30），而10例乳腺增生症患者BRCA1阳性率为90%（9/10）（χ2=14.78，Plt;0.01），在乳腺癌患者中腋窝有淋巴结转移组的表达率明显低于无淋巴结转移组（χ2=15.42，Plt;0.01），BRCA1的表达与肿瘤的组织学类型（χ2=0.156，Pgt;0.05）、肿块大小（χ2=0.587，Pgt;0.05）无关。结论 BRCA1在乳腺癌的发生发展过程中有重要作用，可能成为临床对乳腺癌诊断和判断预后的一个重要指标，可能对乳腺癌的早期诊断及预防有重要意义。 【关键词】 乳腺肿瘤；BRCA1基因；免疫组织化学 ［Abstract］ Objective To study the expression of breast cancer susceptibility gene (BRCA1） in breast cancer and benign tumors of the breast and its clinical significance.Methods Applied ABC immunohistochemical method to detect 52 cases of breast cancer，30 cases of breast fibroadenoma and active in patients with epithelial hyperplasia and 10 cases of breast hyperplasia in the expression of BRCA1.Results The positive rate of BRCA1 in breast cancer was 38% (20/52），which in breast fibroadenoma was 73% (22/30），and 10 cases of breast hyperplasia in patients with BRCA1 positive rate was 90 % (9 / 10） (χ2=14.78，P＜0.01）.The rate in breast cancer with the axillary lymph node metastasis was significantly lower than that of without lymph node metastasis (χ2=15.42，P＜0.01），BRCA1 expression and the organization of tumor types (χ2 = 0.156，P＞0.05），tumor size (χ2= 0.587，P＞0.05） had nothing to do.Conclusion BRCA1 which plays an important role for breast cancer occurring and developing，may become an important indicator for clinical diagnosis of breast cancer and prognosis judgement，which has an important significance to breast cancer prevention and early diagnosis. ［Key words］ breast cancer;BRCA1 gene;immunohistochemistry 乳腺癌是女性最常见的恶性肿瘤之一，近年来已经成为女性因癌症死亡原因之首。它的发生发展与多种因素有关。最近的研究发现，乳腺癌易感基因（BRCA1）的表达下降或失表达对乳腺癌的发生发展具有重要作用［1］。我们采用免疫组化ABC方法检测BRCA1在52例乳腺癌，30例乳腺纤维腺瘤并上皮增生活跃和10例乳腺良性增生组织中的表达，同时分析其与乳腺病临床病理的相关性，探讨BRCA1在乳腺癌发生发展及预后的关系，为乳腺癌的预防、诊断、治疗提供有意义的分子生物学指标。 1 资料与方法 1.1 一般资料 标本取自于广东医学院附属医院和附属福田医院2005～2007年乳腺癌标本，为外科手术切除的女性乳腺病变组织，用10%福尔马林固定
