白藜芦醇治疗大鼠急性胰腺炎肺损伤实验探究 　 作者：任雷，马清涌，王铮，李震东 【关键词】 白藜芦醇；胰腺炎；肺损伤;选择素；大鼠 　　摘要：目的 探讨急性胰腺炎(AP)大鼠肺损伤与肺组织选择素(选择素P、选择素E )表达的关系及白藜芦醇对肺损伤的作用。方法 96只雄性大鼠分为对照组、模型组、治疗组、溶剂组。用40g/L牛黄胆酸钠逆行胆胰管注射建立胰腺炎模型，治疗组在建模后立刻经阴茎静脉注射溶解于吐温80溶剂中的白藜芦醇(10mg/kg)。术后3、6、12h取材，盲法光镜下观察大鼠肺组织病理学改变，同时检测支气管肺泡灌洗液(BALF)中的白细胞数及肺组织髓过氧化物酶(MPO)活性，免疫组化法检测肺脏毛细血管内皮选择素P、选择素E表达。结果 治疗组肺组织损害明显较模型组肺组织轻(Plt;0.05)；随时间推移模型组BALF中白细胞数及肺组织MPO活力逐渐增高，而治疗组BALF中白细胞数及肺组织MPO活力在各时间段均较模型组降低(Plt;0.05)；选择素P、E在模型组呈强阳性表达，治疗组表达较弱。结论 在胰腺炎肺损伤中白藜芦醇可能通过降低肺脏毛细血管内皮选择素表达,使白细胞与内皮细胞黏附减少，炎性细胞局部浸润降低，而达到治疗胰腺炎肺损伤的目的。 　　关键词：白藜芦醇；胰腺炎；肺损伤;选择素；大鼠 　　The experimental study of resveratrol in the acute　pancreatitis combined with lung injury in rats 　　ABSTRACT: Objective To explore the relationship of acute lung injury(ALI) in experimental pancreatitis to selectin P and selectin E and to study the effects of resveratrol on ALI. Methods A total of 96 male SD rats were divided into 4 groups radomly with 24 rats in each group: control group, pancreatitis group (model), resveratroltreated group and tween80treated group (solvent). The model of acute pancreatitis was performed by injecting 4% sodium taurocholate into the biliopancreatic duct. Resveratrol was given through penis vein after modeling. Tween80 was used as a solvent of resveratrol. The rats were killed at 3,6,12 hours after modeling. The degree of ALI was judged by studying the pulmonary morphologic changes, leucocytes of bronchi alveolar lung fluid (BALF) and myeloperoxidase (MPO) in lung tissue. The selectin P and selectin E in vascular endothelium of lung tissue was detected immunohistologically. Results Severe pulmonary morphologic changes were observed in pancreatitis group. The counts of leucocytes in BALF and the amount of MPO were increased with time passing. There was a less degree of ALI in resveratroltreated rats than in pancreatitis rats.
