PCR Technology Overview Introduction PCR principle Setting up a PCR laboratory High-fidelity PCR Touchdown PCR PCR Amplification of Highly GC-rich Regions LA-PCR PCR Primer Design Mutagenic PCR DNA sample preparation RNA sample preparation Strategies for overcoming PCR inhibition RT- PCR Real-time PCR Applications for PCR cloning Introduction What is PCR ? PCR stands for “Polymerase Chain Reaction”, a process to replicate DNA. PCR is a technique used to make numerous copies of [i.e. amplify] a specific segment of DNA. PCR makes it possible to quickly and accurately obtain large quantities of DNA needed in carrying out research in molecular biology, in clinical diagnosis, in criminal investigations requiring forensic analysis, and in virus infectious disease research, e.g. in the ongoing battle against AIDS. Summary Definition Process PCR stands for “Polymerase Chain Reaction”. Objective 用于扩增位于两段已知序列之间的DNA区段。 PCR is to amplify DNA segment whose base sequence located its two ends have been cleared. PCR is a fast and inexpensive technique used to amplify, or make many copies of, small segments of DNA. PCR consists of three steps including _____, ______, ______. Repeated_______, _______and ________cycles multiply the target DNA exponentially. Why shall we study PCR? PCR has become one of the most widely used techniques in molecular biology. PCR has revolutionised molecular biology. PCR makes it possible to quickly and acc
