Abstract：Sodium—coupledneutralaminoacidtransporter3(SNAT3)isamemberofSLC38gene
familywhichprimarilyexpressesinastroeytesinthebrainandretina．IttransportsL—glutaminepre’
dominantlyandplaysanimportantroleinglutamate—glutaminecycleinthebrainandliver．Inorder
toeasilydetecttheexpressionandlocalizationofSNAT3onthemembrane，weconstructedaeukary—
oticexpressionplasmidwithanHA tagattheN-terminusofratSNAT3inthemammalianexpression
vectorpBK-CMVA(1098—1300)byPCRanddoublerestrictionendonucleasedigestiontechniques．
Aftertransientlytransfectedtheplasmidintohumanembryonickidneycells(HEK293T)，expression
ofHA—SNAT3fusion protein wasdetected bywestern bohting．Theresultsuggested thatthe HA-
SNAT3fusionproteincouldnormallyexpressonthemembrane．TheplasmidpBK—CMVA(1098—
1300)一HA—SNAT3willprovideausefultoolfortheinvestigationofsturctureandfunctionofSNAT3
inthefuture．
