APaseActivityinSoybeanandCommonBean asRelatedtoPAvailability TianJiang SouthChinaAgriculturalUniversity,Guangzhou510642,China ABSTRACT Soybean(Glyciuemax伍.)Merr.)andcommonbean(Phaseolus vulgarisL.)aretworepresentativegrainlegumes,whosenutritional valueisuniversallyrecognizedsincetheycontainhighlevelsof proteinandahighproportionofoilandcarbohydrates.Grainlegumes alsoplayaveryimportantroleinsustainableagriculturedevelopment fortheirabiliyttofertilizethesoilbybiologicalnitrogenfixation. However,theproductionofsoybeanandcommonbeanissignificantly limitedbylowphosphorus(P)availabilityinthesoil,especiallyinthe tropicsandsubtropics.Plantshaveevolvedavarietyofstrategiesto adapttolowPavailability.Amongthepossiblemechanisms,acid phosphatases(APases)arebelievedtobeimportantformany physiologicalprocessesrelatedtoPefficiencyinplants.Butstudies aboutthephysiologicalfunctionsofAPasearelimitedandtheresults areconflicting.Inthepresentstudy,systematicfieldnadlaboratory experimentswereconductedtoinvestigatethephysiologicaland biochemicalcharacteristicsofAPasesindifferenttissues/organsas relatedtoPavailabilityinordertogetabetterinsightintotheir physiologicalfunctions.Anappliedcorecollectionofsoybean germplasmwasusedinafieldexperimenttostudythecorrelation betweenleafAPaseactivityandPefficiency.LeafAPasepropertiesas relatedtoPavailabilitywerefurthercharacterizedinanutrient solutionculturewith8soybeangenotypesselectedfromthefieldstudy. Subsequently,APasecharacteristicsintherootsandcoytledonswere studiedintwosoybeangenotypesandtwocommonbeangenotypes contrastinginPefficiencyand/orleafAPaseactivity.Finally,oneof theAPaseisoformswaspurifiedrfomtheleavesofacommonbean genotype,anditsbiochemicalandphysiologicalpropertieswere studied.Ourresultsshowedthat: 1.TherewasagreatgenotypicvariationforleafAPaseactivity amongthetestedsoybeangenotypesint
