A bstract! O bj ective To estab lish a hybridom a cell line secreting m onoclonal antibody (M cAb)
against hum an PSCA. M ethods BALB/c m ouse w as mi m un ized by purified PSCA, A murineM cAb des
ignated D3 w as generated and the specificityw as tested by mi m unoh istochem istry andW estern bloting. Re
sults A positive hybridom e cell lineswhich contained M cAb against PSCA w ere obtained. It belong to IgG1
according to the exam ination of mi munoglobu lin classification. W esternb lot test show ed that theM cAb reac
ted specificallyw ith PSCA w ithout other crossing reactions. The antibody detected PSCA express ion on the
cell surface of norm al and m alignant prostate cells, PSCA protein expression w as iden tified in 38/ 0( 95% )
prmi ary prostate cancers. Therew as no statistical d ifference in PSCA expression level betw een norm al prostate
tissues and BPH, but the PSCA expression level of prostate cancerw as of s ignificant statistical difference.
Conc lu sion s It w as show ed the antibody could be used in d iagnosis and mi m unotherapy of prostate cancer.
