Expressionofhypoxia-induciblefactor-1alpha,hexokinase-Ⅱandlactatedehydrogenase-ⅤinhepatocellularcarcinomaandtheirclinicalsignificanceABSTRACTObjectives:Inthisstudy,weinvestigatedtheexpressionofhypoxia-induciblefactor-1alpha(HIF-1α),asubunitoftranscriptionalfactorHIF-1,inhepatocellularcarcinoma(HCC),paracancerouscirrhosis,noncancerouscirrhosis,andnormalhepatictissues,andthenanalyzeditscorrelationwiththekeyenzymesinglycolysispathway,suchasthehexokinase-Ⅱ(HK-Ⅱ)geneandlactatedehydrogenase-Ⅴ(LDH-Ⅴ)gene.Inaddition,theclinicalsignificanceofthese3geneswerealsoevaluated.Methods:Tumoroustissuespecimenswereobtainedrandomlyfrom56patientswithHCCwhounderwentpartialhepatectomyinourhospital,betweenMarch2007andDecember2011.Noneofthesepatientshadreceivedpreoperativetreatment.Inaddition,surgicallyresectedspecimens,including27adjacentparacancerouscirrhosis,23noncancerouscirrhosis,and11normalhepatictissues,werecollectedascontrolsubjectsduringthesameperiodfromthepatientswhounderwentpartialhepatectomyforvariouscauses,suchasHCCsuperimposedoncirrhosis,portalhypertensionwithhepatitisBvirus(HBV)-relatedcirrhosis,hepatichemangiomaortrauma.Writteninformedconsentwasobtainedfromallpatientsbeforecollectionofspecimens.Thespecimenswerefixedinformalinandthenembeddedinparaffinforimmunohistochemical(IHC)analysis.TheexpressionsofHIF-1α,HK-ⅡandLDH-ⅤgenesproteinweredetectedwithIHCtechniqueusingtheSPstainingassaymethod.TheassessmentofcorrelationbetweenHIF-lαgeneandHK-ⅡorLDH-Ⅴgenewasbasedonthesemiquantitativescaleaccordingtoimageanalysissystem.TheirrelationshipswiththeclinicopathologicalcharacteristicsparametersoftheHCCpatientswerealsoanalyzed.Results:InimmunohistochemicalstainingofHIF-1α,HK-ⅡandLDH-Ⅴgenesprotein,thepositiveexpressionwascommonlydetectedasbuffyoryellowstaining.TheHIF-lαexpressionwasmainlylocalizedinthenucleusandpartiallyinthecytoplasm,whileHK-ⅡandLDH-Ⅴgenesproteinexpressionswerelocalizedinthecytoplasmandevidentlyontheenvelopeofthecells.ThepositiveexpressionrateoftheHIF-1α,HK-ⅡandLDH-Ⅴgenespr
