关键词：人类免疫缺陷病毒1型；细胞毒性T淋巴细胞；高效抗逆转录病毒疗法；HLA-I等位基因TheinfluencialfactorsonHIV-1antigenspecificCTLimmuneresponseMajor:MED.InfectiousDiseaseMasterCandidate:WenZhaoSupervisor:ProfessorYangboTangAbstractCurrentlyacquiredimmunodeficiencysyndrome(AIDS)hasbecomeaseriousinfectiousdiseasesagainstthehealthofallmankind.Highlyactiveantiretroviraltherapy(HAART)couldsuppressthehumanimmunodeficiencyvirus-1(HIV)replication,slowdiseaseprogression,itisthemosteffectivemethodofcontrolandtreatmentofAIDSuptonow.PastresultsshowedthatspecificcytotoxicTlymphocyte(CTL)playakeyroleincontrollingHIV-1replication,butresearchinthisareaismainlyaimedatthosewithoutHAARTtreatmentofHIV-1infectionpersons,andtherelevantHAARTonHIV-1-specificCTLimmuneresponseofthedomesticandinternationalstudieshavereportedlittle,it’sofgreatsignificancetostudyHAARTforspecificCTLimmuneresponsesininfectedpeopleforunderstandingthepost-HAARTimmunefunctionofthebodyscellsandexploretheinternalmechanismofimmunereconstitution.PreviousstudieshaveclearlyindicatedHLA-IallelesinfluenceHIV-1-specificCTLresponsesanddiseaseprogression,However,theresultsoftheseresearchdatasarebasedonnon-Chinesepeoples,theseaspectsofresearchhasnotyetreportedseeinginthedomesticresearch.ThedifferentgroupsofpeopleoftheirHLA-Iallelefrequenciesshowpolymorphism,ChinesepopulationdistributionofHLA-Iallelefrequencywassignificantlydifferentfromotherraces.Therefore,It’sessentialtostudyChinaspopulationtoHLA-IallelefrequencyandthecorrelationofspecificityofCTLimmuneresponse.ItcannotonlyunderstandtherelationbetweenChinainfectedwithHIV-1specificCTLresponsefrequencyandHLA-Ipolymorphism,butalsocanbefoundthespecificHLA-IallelewhichimpactHIV-1-specificCTLresponsesfrequencyanddiseaseprogression.ThisstudychoseChinaGuangdongareaHIV-1infectedpersonsastheresearchobject.FrequencyofInterferon-γ(IFN-γ)secretingcellsinPBMCfrom16untreatedand84HAART-treatedHIV-1-infectedindividualswereassessedbystimulationwithapeptidepoolcontaining12overlappingpeptidesinHIV-1P24,byusingassayofenzyme-linkedimmu
