AbstractCarbofuranisabroad-spectruminsecticidewidelyusedinagriculturetocontrolpestsinhorticulturalcropsfortheproductionandthecontrolofinsect-bornediseases.Theimproperuseandabuseofcarbofuranandtheeffectoffoodchainenrichmentleadtotheseriousresiduseofcarbofuran.Thepresenceofcarbofuraninfoodreceivesworldwideattention.Comparedwithconventionalmethodsfordeterminationofcarbofuran,electrochemicalimmunosensorshavemanyadvantagesincludingsimpleinstrumentation,easyoperation,rapidresponse,highsensitivity,selectivityandhighcompatibilitywithadvancednanotechnologyandmicromachiningtechnologies.Inthisstudy,westudyontheelectrodemodificationandantibodyimmobilizationinordertoselecttheoptimalmethodsforthefabricationofimmunosensor.(1).Thedepositedgoldnanocrystals(DpAu)/4,4’-thiobisbenzenethiol(DMDPSE)multilayers(｛DpAu/DMDPSE｝n)membraneswereusedformodifyingAuelectrodetofabricateamperometricimmunosensor.DpAu/DMDPSEmultilayersweremodifiedbylayer-by-layerself-assemblytechnology.Underoptimizedconditions,thedetectionlimitwas0.06ng/mL.Theadvantagesoftheimmunosensorwereexhibitedinitswiderlinearrange,betterspecificity,reproducibilityandstability.(2).AnamperometricimmunosensormodifiedwithproteinA/depositedgoldnanocrystals(DpAu)wasdevelopedfortheultrasensitivedetectionofcarbofuranresidues.First,DpAuwereelectrodepositedontotheAuelectrodesurfacetoabsorbproteinA(PA)andimprovetheelectrodeconductivity.ThenPAwasdroppedontothesurfaceofDpAufilm,usedforbindingantibody’sFcfragment.Following,antibodywasimmobilizedonthePAmodifiedelectrodeandthenblockedbybovineserumalbumin(BSA).WiththeexcellentelectroconductivityofDpAuandthePA’sorientedimmobilizationofantibodies,thehighlyefficientimmuno-reactionanddetectionsensitivitycouldbeachieved.TheinfluencesoftheelectrodepositiontimeofDpAu,pHofthedetectionsolutionandincubationtimeonthecurrentresponseofthefabricatedimmunosensorwereinvestigated.Underoptimizedconditions,thedetectionlimitwas0.1924ng/mL.(3).Anovelamperometricimmunosensorwasdevelopedforthedetectionofcarbofuran.Fir
