ABSTRACTObjective：Study of fibroblast proliferation and PCNA expression after matrine in glaucoma filtration surgery in rabbit eyes, Preliminary to explore anti-scaring effect of matrine after glaucoma filtration surgery.Methods ： Choose healthy New Zealand white rabbit 40 (male and femaleunlimited), only 1.5 2.0 Kg weight,these rabbit were randomly divided into four groups(each group including 20 eyes of 10 rabbits): group A group B group C groupD. Both eyes in four groups were treated with glaucoma filtration surgery. cotton pieces under the sclera flap were made : 1.0 g/l matrine cotton piece in group A，0.4 g/l matrine cotton piece in group B，0.2 mg/l mitomycin cotton piece in group C, ingroupD. Respectively to observe groups of intraocular pressure, filtering bleb, anterior chamber reaction, complications, and so on in the preoperative and postoperative 1, 3, 7, 14, 28 days. Two rabbits selected randomly in every group were killed on days 1,3,7 ,14 and 28 after surgery.Tissue was harvested from the bleb area and was made pathological section, immunostained and Masson trichromatic dyeing To observe the inflammatory cells, collagen, fibroblasts, the change of muscle fibroblasts and new collagen fibers under light microscope. Two rabbits selected randomly in every group were killed on days 7 after surgery. Remove eye for tem slice, to observe the toxicity of drugs.The data was analyzed statistically by SPSSfor windows.Results：1.IOP:Postoperative 1, 3 days, four groups of IOP was not significantly different (PO.05), and postoperative 7, 14, 28 days ,A, B, C group and D group two two comparison of IOP were significantly different(PO.05), postoperative 7, 14, 28 days ,A, B group had significantly different (PO.05).Filtering blebs: Postoperative 1, 3 days,each group of filtering blebs were diffuse,flat and transparent; postoperative 14d, each group of filtering blebs were bigger than before, diffuse,ridgy and transparent.
