宁夏医科大学硕士学位论文摘要皮性卵巢癌细胞株SKOV3及HO8910细胞内的β-catenin的表达，实现对卵巢癌细胞的增殖作用。关键词上皮性卵巢癌细胞，卵泡刺激素，β-粘连蛋白IIEffectsofFollicleStimulatingHormoneonWnt/β-cateninExpressioninEpithelialOvarianCancerCellABSTRACTObjectiveTodiscusstherelationshipbetweenFSHandWnt/β-catenininEpithelialOvarianCancer（EOC）CellLinesbyexaminingwhetherFSHcaninducetheproliferationofEpithelialOvarianCancer（EOC）CellLinesSKOV3andHO8910andtheexpressionofβ-cateninineachoftheseovariancancercelllinesstimulatedbyFSH.MethodsAfterculturedSKOV3andHO8910beingtreatedbyFSHofdifferentconcentration（20，40，60，80，160mIu/ml）atdifferenttime（24，48，72h）,theabsorbencyofthecellswasdetectedbymethylthiazolyltetrazolium(MTT).TakingFSHofthebestconcentrationasexperimentalgroupandequivalentcellculturemediumascontrolgrouptodetecttheexpressionofβ-cateninineachofthecelllineseffectedbyFSHbyusingimmunofluorescence.Todetecttheexpressionofβ-cateninwithWesternBlotbyrespectivelytakingFSHofdifferentconcentration（20，40，60，80mIu/ml）andequivalentcellculturemediumcultivatedfor48hoursasexperimentalgroupandcontrolgroup.ToconductquantitativeanalysisbyusingQuantity-onetoevaluatethevalueofgrayscaleperformedinproteinband.ResultsFSHcouldinducetheproliferationeffectsinovariancancercelllineSKOV3andHO8910.TheproliferationoftheHO8910cellsstimulatedbyFSHoftheconcentrationsof20，40，60mIu/mlfor48hourswasstatisticallysignifican（tP0.05）.TheproliferationoftheSKOV3cellsstimulatedbyFSHoftheconcentrationof40mIu/mlfor48hourswasthemoststatisticallysignificant（P0.05）.Positiveβ-cateninexpressionwasfoundinthenucleusandcytoplasmofbothSKOV3andHO8910celllines.AfterbeingstimulatedbyFSHfor48hours,thecellsofmeandensitywithpositiveβ-cateninexpressionincreasedsignificantly（P0.05）inSKOV3andHO8910.β-cateninexpressionwasfoundinSKOV3andHO8910celllines.Theβ-cateninexpressionoftheHO8910cellsstimulatedbytheconcentrationsof20,40mIu/mlfor48hoursincreasedsignificantly（P0.05）.Theβ-cateninexpressionoftheSKOV3cellsincreasedsignificantlyonlyafterbeingstimulatedbyaconcentrationof20mIu/mlfor48hours（P0.05）.Con
