第二军医大学硕士学位论文Method: Biological samples are determined by LC-MS/MS after preprocessing. Chromatographic separation was performed on a C18 column（3.0mm×100mm ，3.5μm） with methanol：2mmol·L-1ammonium formate （adjusted to PH3.52 with formic acid）asthe mobile phase.A trip-quadrupole tandem mass spectrum was applied the electrospray ionization（ESI）source and operated in positive ion multiple reaction monitoring mode,SH was detected at m/z 304.2138.1.Six heathy beagle dogs were divided into twogroups,half male and half female. They received oral single dose of SH orally disintegrating microencapsule tablets 0.6mg（test tablet）or SH normal tablets （reference tablet）. Plasma samples collected at designed time .Result:The studies have shown that test tablets have higher Cmax and Tmax thanreference tablets.Those indicate test tablets have characteristics of rapid disintegration.PARTⅢ Research on the tissue distribution of SH in ratsObjective:To determine concentration of SH in rat tissues by LC-MS/MS method,and the characteristics of its tissue distribution were evaluated.Method: Biological samples are determined by LC-MS/MS after preprocessing. The plasma 、heart、liver、brain、spleen、lung and kidney tissues were removed after poured the drug 30mg·kg-1 into the stomach of rats at scheduled time,and weighted those tissues precision by filter paper.Adding two times physiological saline make those tissues intohomogenate,then preserved them in refrigerator under minus eighty degrees .Result:the distribution of SH in rats’ tissues were ClungCspleenCl
