反应停联合他莫昔芬对小鼠乳腺癌术后复发和转移影响 　　基金项目 开封市科技攻关项目(110336) 　　? 　　摘 要 目的：观察反应停联合他莫昔芬对小鼠乳腺癌手术切除后转移和复发的影响。方法：将小鼠4T1乳腺癌细胞植入雌性BALB/c小鼠乳房脂肪垫下。乳腺癌直径达10mm时手术切除，次日开始分别用50mg/kg的反应停和25mg/kg的他莫昔芬单独及联合治疗小鼠3周。治疗效果通过小鼠术后转移和复发率、小鼠血清中Na?+，K?+，Cl?-，glu水平及小鼠存活时间评价。结果：对照组、他莫昔芬组、反应停组、反应停联合他莫昔芬组乳腺癌复发率分别为75%、80%、60%和20%；肺内出现转移灶率分别为80%、100%、50%和25%。与对照组比较，他莫昔芬治疗小鼠血清Cl?-和K?+升高，Na?+和glu降低。反应停及反应停联合他莫昔芬治疗小鼠血清Cl?-、Na?+、K?+和glu与正常小鼠相似。结论：反应停联合他莫昔芬能促进机体电解质平衡，可以用于预防乳腺癌术后转移和复发。? 　　关键词 反应停 他莫昔芬 乳腺癌 肿瘤复发 电解质平衡? 　　doi:10.3969/j.issn.1007-614x.2012.01.002?? 　　Abstract Objective:To investigate the effect of thalidomide combined with tamoxifen on metastasis and recurrence of breast cancer(4T1)after the tumor removal in mice.Methods:4T1 breast cancer cells were injected into the second mammary fat pad of BALB/c mice.The breast cancer was removed by operation when it reached a diameter of 10mm,next day,the treatment was initiated with 25 mg/kg of thalidomide and/or 50mg/kg of tamoxifen for 3w.The therapeutic efficacy was evaluated by the rate of metastasis and recurrence,serum Na?+,K?+,Cl?-,glu and life span in mice.Results:The rates of tumor recurrence were 75%、80%、60% and 20% in the control.tamoxifen.thalidomide and thalidomide combined with tamoxifen groups.respectively.The pulmonary metastasis rates were 80%，100%，50% and 25% in control,tamoxifen,thalidomide and thalidomide combined with tamoxifen groups,respectively.Compared with the control group,serum Cl- and K?+ significantly increased and Na+ and glu decreased in tamoxifen group,whereas Na?+,K?+,Cl?- and glu in thalidomide and thalidomide combined with tamoxifen groups were similar to control group.Conclusion:It is very important for controlling tumor metastasis and recurrence to recover host electrolyte balance after the tumor removal.Thalidomide combined with tamoxifen can promote the host electrolyte balance,which might be used for preventing metastasis and recurrence after breast cancer removal.? 　　Key Words Thalidomide;Tamoxifen;Breast cancer;
