The thesis explores the action mechanisms of near-infrared spectroscopy in the detection of pathogenic bacterias, analyses the near-infrared spectral of pathogenic bacteria cells, taking Salmonella and L.monocytogenes as the research object which is in the logarithmic growth phase, using projection discriminant analysis based on PCA to identification of the pathogen bacteria cells ,as well as the examination limit for a study to get the expected result. 1. Aseptic technique, Research the Growth situation of Salmonella and L.monocytogenes in both Nutrient agar culture medium and Including 0.6% yeast paste pancreas cream peptone soybean agar-agar culture medium(TSA—YE). The results show that，Salmonella and L.monocytogenes are not high to the nutrition demand and Can grow well in both Nutrient agar culture medium and Including 0.6% yeast paste pancreas cream peptone soybean agar-agar culture medium(TSA梇E). 20. Carried out a research into logarithmic growth phase of Salmonella and L.monocytogenes, taking the use of photoelectric turbidimetry. In this study, the growth curves of Salmonella and L.monocytogenes is compared to they growth chart, from which we can see the growth trend of Salmonella and L.monocytogenes so as to determinate the Salmonella and L.monocytogenes logarithmic growth phase.
